Mechanism of action

How Humanin is described to act.

Humanin is a 24-residue peptide encoded by a short open reading frame in the mitochondrial 16S rRNA gene (MT-RNR2) and was the first mitochondrial-derived peptide described. In vitro single-molecule fluorescence and Forster resonance energy transfer experiments in lipid bilayers report that humanin binds Bax and truncated Bid, inhibits Bax self-association and tBid-activated Bax membrane association, and decreases the proportion of Bax present as tetramers, thereby limiting mitochondrial membrane permeability and intrinsic apoptosis. Review literature additionally describes humanin acting extracellularly through cell-surface receptor complexes and reports inhibition of mitochondrial complex I activity with a corresponding reduction in cellular oxidative stress. Humanin expression is reported in several tissues with high metabolic rate and, in brain, in glial cells including astrocytes.

Research findings

In Caenorhabditis elegans, humanin overexpression was reported to increase lifespan in a manner dependent on daf-16/FOXO, and humanin transgenic mice displayed overlapping phenotypes with greater protection against toxic insults. Twice-weekly treatment of middle-aged mice with the potent humanin analogue HNG was reported to alter metabolic healthspan parameters and reduce inflammatory markers. In a rodent surgical menopause model, astrocyte-released humanin was reported to prevent synapse loss in hippocampal neurons. Observational human and cross-species data indicate that circulating humanin generally declines with age, that levels were stable in the naked mole-rat, and that children of centenarians showed higher circulating humanin than age-matched controls. Cardiovascular and energy-metabolism reviews discuss humanin as a candidate marker of mitochondrial function; no controlled human intervention trial of humanin appears in this citation set.
