Mechanism of action

How MOTS-C is described to act.

MOTS-c is a 16-amino-acid peptide encoded by a short open reading frame within the mitochondrial 12S rRNA gene (MT-RNR1), identified as the second described member of the mitochondrial-derived peptide family. In cultured cells and rodent tissue it is reported to inhibit the folate cycle and its tethered de novo purine biosynthesis, producing AICAR accumulation and AMPK activation, with skeletal muscle described as the apparent primary target organ. Under metabolic stress such as glucose restriction, MOTS-c has been shown to translocate from mitochondria to the nucleus in an AMPK-dependent manner and to interact with antioxidant response element (ARE)-regulating stress-responsive transcription factors including NFE2L2/NRF2, a proposed retrograde mitochondrial-to-nuclear signaling route. Separate rodent adipose work attributes thermogenic gene induction to MOTS-c-associated ERK phosphorylation.

Research findings

In the original characterization, MOTS-c administration to mice was reported to prevent age-dependent and high-fat-diet-induced insulin resistance and diet-induced obesity. Follow-up rodent work reported that cold exposure lowered serum MOTS-c and that MOTS-c treatment upregulated brown adipose thermogenic gene expression, increased white adipose browning, and reduced lipid trafficking to the liver. In a transverse aortic constriction mouse model of pressure-overload heart failure, subcutaneous MOTS-c delivered by osmotic pump was associated with attenuated cardiac dysfunction and remodelling, reduced inflammatory response and increased antioxidant capacity alongside AMPK pathway activation, while MOTS-c overexpression reduced H2O2-induced death in cultured H9C2 cardiac cells. Review articles in this set describe endogenous MOTS-c expression as responsive to exercise and metabolic stress, with circulating levels generally lower in aging and metabolic disease. Human data across these citations are observational or expression-level; no controlled clinical efficacy trial of MOTS-c is represented here.
