Mechanism of action

How IGF-1 LR3 is described to act.

Long R3 IGF-I is a recombinant analogue of insulin-like growth factor-I carrying an arginine substitution at residue 3 together with an N-terminal extension peptide, modifications that markedly reduce affinity for the IGF binding proteins while retaining signaling through the type 1 IGF receptor. The reduced binding-protein affinity is the mechanism the literature offers for its greater in vivo potency relative to native IGF-I in several rodent models. Receptor-level work reported that IGFBP-3 inhibits IGF receptor phosphorylation induced by IGF-I, IGF-II, Des(1-3)IGF-I and Long(R3)IGF-I largely by sequestering ligand in the extracellular milieu rather than by interacting with the receptor directly. In intact animals, chronic exposure has been reported to lower circulating growth hormone, IGF-I and IGF binding protein-3, consistent with feedback on the somatotropic axis.

Research findings

In streptozotocin-induced diabetic rats, LR3-IGF-I and des(1-3)IGF-I were reported to be roughly 2.5 to 3 times more potent than native IGF-I at restoring growth rate and nitrogen balance, with the added weight gain composed of protein rather than the fat observed in insulin-treated reference animals. In guinea pigs, seven-day infusion increased the fractional weight of adrenals, gut, kidneys and spleen while lowering plasma IGF-I, IGF-II and IGF binding protein concentrations, without stimulating overall body growth. In finisher pigs the same analogue decreased average daily gain, food intake and plasma IGFBP-3, IGF-I and insulin, and reduced mean and pulsatile growth hormone concentrations, illustrating pronounced species-dependent responses. Protein-engineering work mapped the preferred pepsin cleavage sites within long-R3-IGF-I and generated single-point muteins, one of which showed markedly improved proteolytic stability while retaining growth-promoting activity. A forensic mass spectrometry report identified His-tagged research-grade Long-R3-IGF-I, a form normally produced for biochemical studies, in a confiscated black-market injection vial with the affinity tag still attached.
